چکیده

Interactions of a model surfactant, sodium dodecyl sulfate (SDS), with a water-insoluble model protein, zein, were investigated to gain an understanding of the effects, such as skin irritation and protein denaturation, of surfactants that are common in personal-care products. To elucidate the mechanisms of such effects, the zeinprotein interactionwithSDSinaqueoussolutionswas investigatedusingamultipronged approach involving a range of techniques, such as UV-visible and fluorescence spectroscopy, TOC (total organic carbon analysis), light scattering, and viscosimetry. The zein protein solubilization increases with an increase in the SDS concentration. Solubilization of zein occurs in two distinct stages followed by a complete unfolding of the protein. In the first stage ([SDS] ∼ 4 mM; critical complexation concentration), SDS is incorporated into the globular zein structure, forming small hydrophobic microdomains. From the pyrene fluorescence lifetime decay measurements, the aggregation number of SDS in such hydrophobic microdomains was found to be markedly lower than the aggregation number of pure SDS micelles in the bulk solution. The vibrational fine structure of pyrene fluorescence, however, showed the core of SDS-zein complex micelles to be more hydrophobic than that of the SDS micelles. In the second stage ([SDS] ∼ 200 mM; unfolding concentration), the protein unfolds, as is evidenced by viscosity and dynamic light scattering measurements.

Significant effect of polar head group of surfactants on the solubilization of Zein in mixed micellar (SDS-DDAB) media.

In recent years, the ability of ionic surfactants such as sodium dodecyl sulfate (SDS) to solubilize the water-insoluble protein Zein in aqueous solution, referred to as the Zein solubilization test, has been investigated to rank the skin irritation potential and protein denaturation potential of surfactants. However, the potential of cationic surfactants towards solubilization of Zein has not ...

Zein protein interactions, rather than the asymmetric distribution of zein mRNAs on endoplasmic reticulum membranes, influence protein body formation in maize endosperm.

Prolamin-containing protein bodies in maize endosperm are composed of four different polypeptides, the alpha-, beta-, gamma-, and delta-zeins. The spatial organization of zeins within the protein body, as well as interactions between them, suggests that the localized synthesis of gamma-zeins could initiate and target protein body formation at specific regions of the rough endoplasmic reticulum....

The Interaction between Zein and Lecithin in Ethanol-Water Solution and Characterization of Zein–Lecithin Composite Colloidal Nanoparticles

Lecithin, a naturally small molecular surfactant, which is widely used in the food industry, can delay aging, enhance memory, prevent and treat diabetes. The interaction between zein and soy lecithin with different mass ratios (20:1, 10:1, 5:1, 3:1, 2:1, 1:1 and 1:2) in ethanol-water solution and characterisation of zein and lecithin composite colloidal nanoparticles prepared by antisolvent co-...

The Maize y-Zein Sequesters a-Zein and Stabilizes Its Accumulation in Protein Bodíes of Transgenic Tobacco Endosperm

Zeins are seed storage proteins that form accretions called protein bodies in the rough endoplasmic reticulum of maize endosperm cells. Four types of zeins, a, p, y , and 6, aggregate in a distinctive spatial pattern within the protein body. We created transgenic tobacco plants expressing a-zein, y-zein, or both to examine the interactions between these proteins leading to the formation of prot...

Role of non-covalent interactions in the production of visco-elastic material from zein.

The role of non-covalent interactions in the formation of visco-elastic material from zein was investigated. Hydrophobic interactions were evaluated through the addition of various salts from the Hofmeister series. Urea, ethanol, and beta mercaptoethanol (β-ME) were used to evaluate the effects of protein denaturation and disulfide bonds on zein's ability to form a visco-elastic material. The a...
